For a brand new account with very little unlocked, what professions and builds would be able to get up to speed (intended) quickly to open up the maps all over for GotT?

In short, it boils down to finishing the campaigns. Assassin would be the profession of choice, but given the lack of difficulty in NM and the number of hours difference that this will make, other professions could also be chosen. It should be noted that the heroes who accompany him, the builds, i.e. the skills unlocked, and even the mercenary heroes unlocked or not, will make a noticeable difference.
It would be easier to run a character from your new account with an account that is already complete or semi-complete.

In addition to what Neutral said above... you could have alook at the beginner heroes guide on pvx - following it gets you to a self sufficient set up very quickly.
